An enzyme assay was developed for studies on peroxidase activities in humic lake water. 3,4-Dimethoxybenzyl alcohol (veratryl alcohol, VeraOH) was used as tracer substrate, and peroxidase (EC 1.11.1.7) activity was measured by high-performance liquid chromatography. The chemical stability of VeraOH and its application as peroxidase substrate was tested under light and dark conditions, different hydrogen peroxide (H2O2) concentrations and humic matter contents. VeraOH was stable under low UV radiation at in situ conditions in lake water (<0.010...0.25 kJ m–2 d–1), laboratory conditions (<0.05...0.30 kJ m–2 d–1), and low (1...100 μM) H2O2 concentrations. However, peroxides oxidized VeraOH above 1...10 mM H2O2 concentration in sterile Millipore-Q and humic lake water. Dark incubations showed little VeraOH oxidation products. The developed peroxidase assay was tested in the growth medium of Phanerochaete chrysosporium and a bacteria isolate (P.M.D. 20.4.3.1) from mesohumic lake Pääjärvi. Peroxidase activities were also measured in natural microbial communities under standard laboratory and under in situ conditions in humic lake water. Incubation times of about 5 to 12 days were usually needed to record significant (P < 0.05) peroxidase activities, in lake waters. In situ peroxidase activities varied in pelagial surface water (0...0.5 m) on a seasonal scale between 74 nmol L–1 h–1 and 273 nmol L–1 (mean: 176 nmol L–1 h–1) and within the water column between 110 nmol L–1 h–1 and 800 nmol L–1 h–1 (mean: 500 nmol L–1 h–1) in polyhumic lake Mekkojärvi. 相似文献

Thirty four elements dissolved in water and 33 elements bound in particulate matter were analysed in the small river Saale (Thuringia, Germany, MQ=23 m3/s) in 1996 and the results were compared with those obtained in 1950 (44 elements). Monthly sampling was used to eliminate fluctuations caused by seasons and weather. Comparison of the element contents of a river over a 40-year-time span provides interesting insight into the anthropogenic change in the catchment area of the river with regard to „global change”.Without taking into consideration systematic errors, the analysed elements can be divided into three groups:
- (a) Elements whose average annual analytical values in 1996 were lower by >50% than those in 1950: Al, As, Ba, Fe, Pb, Zn (in solution), and As, B, Cr, Li, Mn, Pb, Se, Sn, U (suspended).
- (b) Elements whose average annual analytical values in 1996 are in the range of those in 1950, i.e., are within 50–150%: Ca, Co, Cr, F, K, Li, Mn, Na, Si, Sr, Ti, U (in solution), and Hg, Cr, Cu, Sc, Sr, Ti, Zn (suspended).
- (c) Elements whose average annual analytical values in 1996 were higher by >150% than those in 1950: B, Cd, Mg, Ni, Rb, Sc (in solution), and Ba, Ni, P, Zr (suspended).

Effects of eruption history and cooling rate on lava dome growth 总被引:1,自引:1,他引:0
To better understand the factors controlling the shapes of lava domes, laboratory simulations, measurements from active and prehistoric flows and dimensional analysis were used to explore how effusion history and cooling rate affect the final geometry of a dome. Fifty experiments were conducted in which a fixed volume of polyethylene glycol wax was injected into a tank of cold sucrose solution, either as one continuous event or as a series of shorter pulses separated by repose periods. When the wax cooling rates exceeded a critical minimum value, the dome aspect ratios (height/diameter) increased steadily with erupted volume over the course of a single experiment and the rate at which height increased with volume depended linearly on the time-averaged effusion rate. Thus the average effusion rate could be estimated from observations of how the dome shape changed with time. Our experimental results and dimensional analyses were compared with several groups of natural lava flows: the recently emplaced Mount St Helens and Soufrière domes, which had been carefully monitored while active; three sets of prehistoric rhyolite domes that varied in eruptive style and shape; and two sets of Holocene domes with similar shapes, but different compositions. Geometric measurements suggest that dome morphology can be directly correlated with effusion rate for domes of similar composition from the same locality, and that shape alone can be related to a dimensionless number comparing effusion rate and cooling rate. Extrapolation to the venusian pancake domes suggests that they formed from relatively viscous lavas extruded either episodically or at average effusion rates low enough to allow solidified surface crust to exert a dominating influence on the final morphology. 相似文献

Suspended particles are instrumental in controlling the reactivity, transport and biological impacts of substances in aquatic environments, and provide a crucial link for chemical constituents between the water column, bed sediment and food chain. This article reviews the role of suspended particles in the chemical and biological cycling of trace constituents (trace metals, organo-metallic compounds and hydrophobic organic micropollutants; HOMs) in estuaries, with particular emphasis on the effects of and changes to particle reactivity and composition.The partitioning (or distribution coefficient,KD ) and bioavailability of chemical constituents, and assimilation efficiency (AE) of such by bivalve suspension feeders, are identified as key parameters requiring definition for accurate biogeochemical modelling, and the discussion centres around the determination of and controls on these parameters. Particle-water interactions encompass a variety of physical, biological, electrostatic and hydrophobic effects, and are largely dependent on the character and concentration of suspended particles and salinity. The salinity-dependence results from the competing and complexing effects of seawater ions for trace metals, and the compression of water in the presence of dissolved seawater ions and consequent salting out of neutral solute (HOMs, organo-metallic compounds and some trace metal complexes). The extent of biological solubilization of chemical constituents from suspended particles is dependent on the nature of chemical components of the gastro-intestinal environment and their interactions with ingested particles, and the physiological (e.g. gut passage time) and chemical (e.g. redox conditions and pH) constraints imposed on these interactions. Generally, chemicals that associate with fine, organic-rich particles (or, for some HOMs, fine inorganic particles), and desorb at pH 5–6 and/or complex with digestive enzymes or surfactants are most readily solubilized in the gut. The extent of assimilation of solubilized chemical is then determined by its ability to pass the gut lining and partition into cytosolic material.In practice, KD and AE are determined experimentally by means of radiotracers added to contained suspensions or mesocosms, while operational measurement of bioavailability relies on in vitro chemical or biological (enzymatic) extraction of particles. What is lacking, however, and is identified as an ultimate goal of future research, is the ability to predict these parameters from theoretical principles and thermodynamic constants. Since many of the inherent interactions and mechanisms are controlled by particle composition and reactivity, a more immediate objective would be better characterization of the biogeochemical properties of suspended particles themselves. This includes chemical resolution of the bulk organic matter, definition of the abundance and synergistic effects of component sorbent phases, and determination of the effects of particle-seawater ion interactions on the reactivity of the particle surface. 相似文献

Abstract. Animal-animal interactions are recognized as critical in benthic invertebrate recruitment. The hypothesis tested was that benthic recruitment is more successful in low densities of infaunal suspension feeders than in high densities. Densities of young hard clams Mercenaria mercenaria (L.) ranging from 82.5 to 330m-2 were placed in defaunated boxes of sand. The mean number of species, mean number of individuals, mean wet weight biomass, mean species richness and mean dominance index per sample were calculated per density of hard clams from May to October. The experiment did not support the hypothesis. It was concluded that feeding behaviour of dense populations of M. mercenaria did not preclude successful recruitment of other benthic species. 相似文献
